Detection of aminophylline in serum using an immunochromatographic strip test

Abstract: Aminophylline (AMP), a bronchodilator drug, is a modified form of theophylline (the pharmacological active ingredient). AMP is often given to treat respiratory diseases, including bronchial asthma, blocked emphysema, panting bronchitis, and cardiogenic asthma. In this study, we developed a rapid and sensitive method to determine AMP concentration in mouse serum. A sensitive monoclonal antibody against AMP was produced by immunizing BALB/c mice with a well-characterized AMP2-keyhole limpet hemocyanin and coatin… Show more

“…Refer to previous methods in the laboratory to label the mAbs with AuNPs (Chen et al, 2016;Guo et al, 2019;Wu et al, 2020) and in the first step, the gold nanoparticles GNPs are prepared. Briefly, 50 mL of HAuCl 4 • 4H 2 O (0.01%, m/v) solution was heated to boiling with constant stirring.…”

“…Using chromatographic methods, multiresidue analytics with detection limits lower than 1 ng•mL −1 for many antibiotics can be performed [21]. However, this method is restricted to analytical laboratories due to the need for highly trained staff and cost-intensive laboratory-based equipment [15,16,22]. Nowadays, immunological tests such as ELISA or lateral flow assays (LFAs) as well as microbial test kits are commonly used for monitoring of milk samples [7,15,17].…”

“…Nowadays, immunological tests such as ELISA or lateral flow assays (LFAs) as well as microbial test kits are commonly used for monitoring of milk samples [7,15,17]. LFAs are simple, easy-to-handle and are usually performed within minutes, which makes them easy to use, even for untrained personnel [22]. However, such LFAs lack sensitivity and a quantitative measurement is typically not possible.…”

“…Immunoassays based on antibodies and antigens are potentially sensitive methods of TZP. The enzyme-linked immunosorbent assay (ELISA) has been used for TZP detection (Gui et al, 2006;Hong et al, 2018), which is highly specific and require minimum sample preparation, is effective high throughput applications; however, the assay is time consuming (Wu et al, 2020;Xu et al, 2020;Zhang et al, 2019). The lateral flow immunochromatography assay (LFA) is easy, fast (5-10 min), and sensitive (Kong et al, 2019;Li et al, 2020;Wang et al, 2020;Zeng et al, 2020).…”
