Detection of Bacterial Alkaline Phosphatase Activity by Enzymatic In Situ Self-Assembly of the AIEgen-Peptide Conjugate

Abstract: Abnormal levels of alkaline phosphatase (ALP) activity are associated with various diseases, and many ALP probes have been developed to date. However, the development of ALPsensitive probes for living cells, especially for the detection of bacterial ALP, remains challenging because of the complex and dynamic context. In this study, we constructed the first fluorescent probe (TPEPy-pY) for sensing bacterial ALP activity. TPEPy-pY is an AIEgen-peptide conjugate with property of aggregation-induced emission (AIE)… Show more

“…In addition, they offer a complementary strategy to smart wound technologies and point of care (PoC) devices [11] . Current small‐molecule fluorescent probes utilise enzyme‐based biomarkers to facilitate the detection of pathogenic bacteria, [12] which include elastases, [10e] phosphatases, [13] glycosidases, [14] proteases [15] and lipases [16]…”

“…In another study, Yang et al developed an alkaline phosphatase (ALP)-activated fluorescent turn-on AIEgen-peptide conjugate probe 24 (Figure 12A) (Zhang et al, 2020b). Probe 24 can undergo dephosphorylation by bacterial ALP to form an aggregated fibrous structure on the bacterial surface and provide AIE-based fluorescence.…”
