Detection of Clostridium tetani in human clinical samples using tetX specific primers targeting the neurotoxin

Abstract: Tetanus resulting from ear injury remains an important health problem, particularly in the developing world. We report the successful detection of Clostridium tetani using tetX specific primers targeting the Cl. tetani neurotoxin. The sample was obtained from an ear discharge of a case of otogenic tetanus in a 2-year-old male child. Based on the culture results of the ear discharge, Gram staining and virulence testing by genotyping, a diagnosis of tetanus was confirmed. This is the first report from India on t… Show more

“…Bacterial DNA is commonly extracted from enrichment broth cultures or colonies on agar plates with commercial DNA extraction kits. 2,25,[41][42][43] In conventional PCR, the readout is performed by electrophoresis of DNA on an agar gel stained with ethidium bromide. More accurately, the sequencing of PCR products gives unambiguous results.…”

“…However, C. tetani are difficult to isolate due to the characteristics of anaerobic bacteria (12,13). Recently, the detection of tetanospasmin in C. tetani using real-time polymerase chain reaction (PCR) has become possible, and it has been used to diagnose tetanus in livestock (14)(15)(16).…”

“…Heavy-metal ions and organic pollutants such as pesticides and toxins accumulate at the top of the biological chain, threatening public health and inducing physiological defect to infants . However, bacteria and viruses such as Escherichia coli, Staphylococcus aureus, Clostridium tetani, human immunodeficiency virus (HIV), and hepatitis B virus (HBV) also cause severe health problems. Associated with the pathological changes of the human body, the amounts of various biomarkers also varies to abnormal levels .…”
