Detection of HHV‐8 (human herpesvirus‐8) genomes in induced peripheral blood mononuclear cells (PBMCs) from US blood donors

Abstract: HHV-8 genomes were not detected from PBMCs, induced/cultured PBMCs and CD19+ B cells from 164 blood donors. The level of detectable HHV-8 genomes in blood donors seems to be extremely low, if they exist.

“…More recently, Qu et al [6], testing purified DNA equivalent to more than 3×10 4 B-cells from 861 donors, including 40 samples from HHV-8 seropositive individuals, did not detect HHV-8 DNA in any of these samples. Furthermore, HHV-8 genomes were not detected from induced/cultured PBMCs and CD19+ B-cells from 164 blood donors, including seven HHV-8 seropositive samples [24]. Unfortunately, in our work, we could not isolate PBMCs from whole blood samples, as repository samples were immediately frozen after whole blood collection at the blood bank.…”

“…A third study induced peripheral blood mononuclear cells and CD19+ B cells from 164 blood donors and no HHV-8 was detected indicating the level of HHV-8 in blood donors is extremely low, if present at all. 11 Limitations of our study include small sample size and the inability to measure efficacy of leukoreduction filtration on blood from healthier study subjects without KS since none of them had detectable HHV-8 DNA.…”

“…For the in vitro experiments, a total of 5×10 5 B cells and 500 µl of culture supernatant were collected post-HHV-8 exposure, pelleted, and assayed by PCR (50, 51). Samples were treated with 1 µl DNase in 10 µl buffer (Sigma) for 15 min and then lysed in easyMAG buffer (NucliSENS).…”

“…Phocine herpesvirus was added as an internal control (52). DNA was mixed with a primer set specific for HHV-8 K8.1 (51), and the real-time PCR was done using a 7000, 7500, or ViiA7 ABI system (Applied Biosystems). B cells isolated from PBMC obtained from KS cases and controls were treated and tested for HHV-8 DNA as described above.…”
