Detection of human neutrophil elastase by aptamer affinity capillary electrophoresis coupled with laser-induced fluorescence using specified site fluorescently labeled aptamer

Abstract: As a multifunctional serine protease, human neutrophil elastase (HNE) plays critical roles in a variety of physiopathological processes, such as acute lung injury, emphysema, atherosclerosis, and arthritis. The quantification of HNE is important in many applications. In this paper, we report an aptamer affinity capillary electrophoresis coupled with laser-induced fluorescence (CE-LIF) assay for detection of HNE using a tetramethylrhodamine (TMR)-labeled DNA aptamer probe. The affinity complex of HNE and DNA ap… Show more

“…Aptamers are obtained from the random oligonucleotide libraries through the systematic evolution of ligands by the exponential enrichment (SELEX) process including the repetitive processes of binding, selection, and amplification of oligonucleotides (Bai et al, 2017; Bostan et al, 2017; M. Jafari et al, 2018; Khoshbin, Housaindokht, Izadyar, Verdian, et al, 2019; S.‐E. Wang & Si, 2013).…”

“…Aptamers conjugated to gold nanoparticles were employed in a CE-based chemiluminescence immunoassay for the analysis of thrombin in serum [40]. In addition, a CE assay with laser-induced fluorescence detection used aptamers to measure the protease human neutrophil elastase in serum [41].…”

“…The development of the aptamer‐based ACE method requires the optimization of several parameters: (i) composition of the BGE, (ii) ion presence (Mg 2+ and K + are important for the aptamer's activity and stability), and (iii) pH and temperature . These methods were used for the detection of several proteins (thrombin, human neutrophil elastase, HIV‐1 reverse transcriptase) , hormones , immunoglobulin E , alkaloids or tumor markers , and even small molecules. Moreover, several research groups used the aptamer‐based ACE technique with nontraditional aptamers for binding studies.…”

“…For example, G‐protein‐BODIPY FL GTPγS, hairpin‐structured DNA‐peptide nucleic acid, concanavalin A‐monosaccharides, dorzolamide‐human carbonicanhydrase II, and tubulin‐anti‐mitotic compounds pairs were examined . Other examples of the aptamer‐based ACE applications are summarized in Table .…”
