Detection of malignant haematopoietic cells in the cerebrospinal fluid by conventional cytology and flow cytometry

Abstract: Morphological evaluation of cerebrospinal fluid (CSF) samples by light microscopy is the method of choice for the detection of central nervous system involvement in patients with haematological malignancies. In this retrospective study, we assessed the value of three-colour immunophenotyping by flow cytometry in addition to conventional cytology in 28 patients with and 17 patients without pre-existent haematological malignancy in whom CSF analysis was performed because of neurological abnormalities (n = 37) or… Show more

“…Previous studies have reported a significant increase of 20–600% in the detection of malignancy by FC [5-7, 11, 14]. In our study, the rate of positivity for malignant cells was increased by 60% by FC, similar to most of the previous reports.…”

“…Several studies indeed underline the importance of multiparameter flow cytometric immunophenotyping for efficient and reliable diagnosis versus exclusion of CNS lymphoma or IOL localizations. 87, 88, 89, 90, 100, 101, 102, 103, 104 For CSF analysis multiple 3-, 4-, 6- or even 8-color single-tube screening panels and protocols have been described (see Kraan et al 106 for an extensive review and description of labelings and methods). In this section we discuss the design and evaluation of the 8-color EuroFlow SST and related sample preparation protocols aimed to evaluate CSF and vitreous biopsy samples, in patients suspected of lymphoma.…”

“…[66][67][68][69][70][71] The advantages of CSF flow cytometry are that it requires small volumes of CSF (<2 ml), is rapidly available, and quantitates tumor volume in CSF.
