“…The method of single cell gel electrophoresis (SCGE; comet assay) according to [9] was performed with minor modifications suggested in [10,11]. Briefly: 2.5 x 10 4 AT-MSCs or CDy-AT-MSCs in 85μl of 0.75% low melting point agarose in PBS were spread on a base layer of 100 μl 1% normal-melting point agarose in PBS, placed on microscope slides and covered with a cover slip.…”

“…We evaluated DNA-damaging (genotoxic) and DNA-protective effects of 24-h treatment of HepG2 and Caco-2 cells with five different compounds of essential oils: eucalyptol (0-5000 µM), eugenol (0-600 µM), borneol (0-1500 µM), carvacrol (0-300 µM) and thymol (0-200 µM). The level of DNA lesions was measured by standard alkaline single cell gel electrophoresis (SCGE; comet assay) suggested by Singh et al [60] and modified by Slamenova et al [61] and Gabelova et al [62]. At concentrations ≤IC 50 the compounds studied did not induce DNA strand breaks either in human HepG2 or in Caco-2 cells, however, incubation of human cells in the presence of the monoterpens thymol and carvacrol led to a significant protection of cellular DNA against a strong oxidative agent -hydrogen peroxide [47].…”

“…45 A relationship between the significantly increased frequency of chromosome aberrations (CA) and cancer risk was demonstrated in spite of CA unspecificity. 46 Studies proved the Comet assay to be more sensitive than CA 47 and a very strong association was shown between the two types of damage, 47,48 suggesting that the Comet assay responses may clearly predict cytogenetic damage.…”
