Immunological functions of the pharyngeal tonsil, palatine tonsils and blood leucocytes of children undergoing tonsillectomy were evaluated by determining T or B lymphocytes, the response to mitogens, and the cell-mediated immunological responses to tuberculin. In all the test systems used similar results were obtained with cells derived from either the palatine or pharyngeal tonsils. The mean percentage of T lymphocytes was significantly higher in the peripheral blood than in tonsils, but the reverse was true of B lymphocytes. The reaction to PHA was lower in tonsillar cell culture than in blood cell culture, but tonsillar cells reacted better to Con A than blood cells. In lymphocyte transformation tests tonsillar cells reacted to specific antigen (tuberculin) and this reaction was significantly higher than that of the parallelly tested blood lymphocytes. Further, in about 50% of the children tested, tuberculin caused migration inhibition of the mixture containing tonsillar cells and guinea pig peritoneal cells. Surprisingly, nearly identical results were obtained if migration inhibition test was performed with tonsillar cells alone. Consequently, poorly migrating tonsillar cells are nevertheless usable for direct migration inhibition testing.

Virus-specific cell-mediated immunity, as determined by in vitro lymphocyte transformation (LT), was demonstrated in calves following infection and vaccination with respiratory syncytial virus (RSV). After experimental infection, 4 of 6 gnotobiotic calves and 6 of 21 conventional calves developed a significant LT response to RSV. By means of a whole blood assay, the LT responses of calves were examined after vaccination with an inactivated vaccine, which consisted of glutaraldehyde-fixed bovine nasal mucosa cells persistently infected with a bovine strain of RSV (GC), a live modified bovine strain of RSV (MV), or a live temperature-sensitive mutant of a human strain of RSV (ts-l). Three weeks after vaccination, a virus-specific LT response was detected in 6 of 6 calves given the GC vaccine, 0 of 4 calves given the MV vaccine, and 2 of 4 calves given the ts-l vaccine. The magnitude of the response was greatest in those animals given the GC vaccine. There was no significant correlation between the magnitude of the LT response and levels of serum neutralising antibody. However, the LT response did correlate with serum antibody measured by the single radial haemolysis test 3 weeks after the first vaccination. LT activity to RSV was associated with T and not B lymphocytes. The development of a virus-specific LT response in calves given an inactivated RSV vaccine was not associated with an increase in respiratory disease following challenge with live virus, but rather was related to increased resistance to RSV infection.

Samples of colostrum, maternal blood, and cord blood from a group of 21 women were examined for the presence of cellular reactivity to respiratory syncytial (RS) virus using a transformation assay and for the level of specific IgA, and IgG, and IgM antibodies to RS virus by membrane immunofluorescence. Six of the 18 colostral cell cultures and six of the 16 maternal blood cultures gave a significant proliferative response to RS virus antigen, although a positive response in both local and systemic cell cultures was found in only one another. In addition, one of 18 samples of cord blood gave a proliferative response to RS virus antigen. Detectable titres of IgA antibodies to RS virus were found in 15 of the 20 samples of colostral whey and in 13 of the 17 samples of maternal plasma examined. RS virus-specific IgG antibodies were detected in 10 of 20 colostral whey samples and in all samples of maternal cord plasma. In this study, it was not possible to demonstrate a relationship between a positive proliferative response of colostral cell cultures to RS virus and the level of specific IgA and IgG antibodies in colostral whey. Similarly, the proliferative response of material blood cultures was unrelated to the titre of specific IgA or IgG antibodies in maternal plasma. The relevance of the local cellular proliferative response to RS virus in colostral cell cultures to the protection afforded by breast-feeding is discussed.
