Determination and pharmacokinetic study of dauricine in rat plasma by UPLC–MS/MS

Abstract: Dauricine is the major bioactive component isolated from the roots of Menispermum dauricum D.C., a bisbenzylisoquinoline alkaloid derivative, and has shown multiple pharmacological properties. In this work, a sensitive and selective ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed for determination of dauricine in rat plasma and its application to pharmacokinetic study of dauricine after intravenous and oral administration in rats. After addition of daurisoline… Show more

“…Liquid chromatography conditions separate isocorynoxeine from the endogenous interfering substances as much as possible from the analyte retention time [29][30][31][32][33][34]. In the investigation of chromatographic conditions, mobile phase systems such as methanol-water, acetonitrile-water, methanol-0.1% formic acid, and acetonitrile-0.1% formic acid were investigated.…”

“…Optimum liquid chromatography conditions separate the endogenous interfering substances from the analyte and internal standard retention times as far as possible (Ma et al, ; Jin et al, ; Wang et al, 2017; Geng et al, ). We tried acetonitrile–0.1% formic acid, acetonitrile–10 mmol/L ammonium acetate (containing 0.1% formic acid), methanol–0.1% formic acid and methanol–10 mmol/L ammonium acetate (containing 0.1% formic acid), using gradient elution.…”

“…The choice of positive and negative electrodes for electrospray ESI is often evaluated in methodology [13, 14]. As an alkaloid and alkaline compound, delavinone is more suitable for ESI positive electrode detection.…”
