Determination of action spectrum for sparfloxacin‐photosensitized single‐strand breaks in plasmid pBR322 DNA

Abstract: The findings suggest that protection of skin from short wavelengths of visible light beyond the absorption wavelengths as well as UVA light is of primary importance in prevention against induction of SPFX phototoxicity.

As observed for the other FQs DNA targets, most of 6,8 dihalogenated FQs (LOM, FLE, ORB, BAY and SPA) show a higher potential as DNA cleaving agents; also in this case methoxy substitution (MOX) tends to reduce this effect (40,45).…”

“…Nonetheless, although ocular (cornea) phototoxicity was observed in LE rats treated with 8-MOP, the detection of retinal phototoxicity from systemic drugs that absorb light above 400 nm requires use of a visible light source. Because SPFX has an absorption band in the range of 320 to 450 nm, 19 the lack of retinal phototoxicity after treatments with SPFX in this study may relate to the use of a UV source. 8-Methosypsoralen, LMFX, and PFD do not absorb visible light.…”

“…Numerous FQs have been reported to induce SSB on plasmid DNA (7,11,14,16–32); the results are shown in Table 2. As a general trend, FQs with a fluorine at C‐8 (LOM, FLE, ORB, BAY and SPA) show a higher potential as DNA cleaving agents, while methoxy substitution (MOX) tends to reduce this effect.…”
