A high performance capillary electrophoresis (HPCE) method was presented to identify and quantitate free amino acids during fermentation by Bacillus subtilis. Amino acids, pre-column derivatized with phenylisothicyanate, were separated and characterized by HPCE. In order to optimize separation conditions, the assay was developed by varying the β-cyclodextrin concentration and pH of the background electrolyte. A buffer system comprising 30 mM phosphate and 3 mM β-cyclodextrin at pH 7.0, voltage of 20 kV and detection wavelength of 254 nm showed the best results, with 17 out of 20 phenylthioncarbamyl amino acids in a solution adequately separated. For quantification, p-aminobenzoic acid was added as an internal standard. Analysis of free amino acids in Bacillus subtilis culture medium using this method revealed good consistency with the values obtained using conventional ninhydrin-based amino acid analyzer. Four free amino acids (aspartic acid, glutamic acid, proline, and tyrosine) concentration in an extracellular matrix during fermentation by Bacillus subtilis were mainly monitored using this method.

The MEKC method is described for the quantitative analysis of 17 amino acids (AA) in pharmaceutical products. The method is based on simply derivatizing the AA with (2-naphthoxy)acetyl chloride under mild conditions. The resulting derivatives were separated by MEKC with borate buffer (35 mM; pH 9.50) including 150 mM SDS at the applied voltage of 25 kV in an uncoated capillary (effective length, 40 cm) and monitored by UV at 230 nm. The detection limits of the amino acid derivatives were in the range of 3.0-8.0 microM (S/N = 3, injection 5.0 s, 6 895 Pa). The precision (RSD) and accuracy (relative error) of the method for intra- and interday analyses of the analytes are all below 5.2%. The amino acid derivatives are stable at room temperature for 33 h studied and the molar absorptivity of the alanine derivative (used as a model) is stable over a wide pH range of 3.00-12.00. This is favorable for monitoring the derivatives in various pH by CE or LC. Application of the method to the analysis of multiple AA in a liquid injection formulation proved satisfactory.

This paper describes a number of articles that have been published on amino acid analysis using CE during the period from June 2003 to May 2005. This review article follows the previous ones of Smith (Electrophoresis 1999, 20, 3078-3083), Prata et al. (Electrophoresis 2001, 22, 4129-4138), and Poinsot et al. (Electrophoresis 2003, 24, 4047-4062). Several new developments in amino acid analysis with CE are reported concerning UV detection, LIF, MS, and NMR. In addition, we describe articles concerning clinical and pharmaceutical studies, neuroclinical applications, and agricultural and food analysis.
