Determination of Azide in Blood and Urine by Gas Chromatography-Mass Spectrometry

Abstract: A sensitive and simple method for determining azide in blood and urine using an extractive alkylation technique was devised. This inorganic anion was alkylated with pentafluorobenzyl bromide using tetradecyldimethylbenzylammonium chloride as the phase-transfer catalyst. 1,3,5-Tribromobenzene was used as an internal standard. The obtained derivative was analyzed by gas chromatography-mass spectrometry using the negative ion chemical ionization mode with isobutane as the reagent gas. The calibration curves for a… Show more

“…"Current sodium azide detection methods include chromatography [9][10][11] and electrochemical detection, [12][13][14] which involves complicated procedures and specialized instruments," said Professor Wang. “…Do analizy azydku sodu stosowano także chromatografię cieczową [13,14,22] lub jonową [23]. Współcześnie metodą z wyboru jest chromatografia gazowa sprzężona ze spektrometrią mas oraz procesem izolacji z matrycy biologicznej i derywatyzacją [2,24].…”

“…Sodium azide also used to be analyzed by liquid chromatography [13,14,22] or ion chromatography [23]. At present, the method of choice is gas chromatography-mass spectrometry (GC-MS) coupled with the process of extraction from biological material and derivatization [2,24].…”

“…The specificity of mass spectrometry (MS) is much better than those of the other methods mentioned above, since MS can determine the analyte using its mass to charge ratio. Since N 3 -is a nonvolatile molecule, GC-MS can be used after derivatizing N 3 -to a volatile pentafluorobenzyl (PFB)-N 3 [11,12]. Determining N 3 -by GC-MS has provided the best specificity as well as sensitivity achieved thus far, leading to a limit of detection (LOD) of 2.4×10 -7 M [12].…”

“…The precursor ion Au(N 3 ) 2 -was formed and extracted with octanol within a few minutes-much shorter than the 30 min required to derivatize N 3 - [11,12]. Extraction with octanol allowed us to eliminate any biological substances that were dissolved in the water, and MS/MS eliminates more interfering ions than single MS as m/z values are selected twice.…”
