Determination of Bone Developments of Rat Anterior and Posterior Extremity Bones in Prenatal and Postnatal Period by Double Staining Method

Abstract

Objective: Detection of the morphological development of the bones of the anterior and posterior extremities by staining the rat fetus and newborn skeleton with double staining method.
 Methodology: In the study, a total of 7 groups (3 of them were prenatal and 4 of them were postpartum) were formed from the fetuses and offsprings obtained from 13 pregnant rats. The anterior and posterior extremity images of fetuses and offsprings were examined under a stereo stereomicroscope, and ossification findings were determined and total bone length, ossification length and ossification areas were calculated in the ImageJ program.
 Results: The first ossification center; It was seen on the 18th day of pregnancy in the clavicula, scapula and humerus and on the 20th day of pregnancy in the radius, ulna, femur, tibia, fibula and 2-5 metatarsal bones. Secondary centers of ossification were seen at day 0 in the scapula and humerus, at day 7 postpartum in the ulna and radius, and at day 12 postnatal in the femur and tibia. In the study, the ossification rate in 20-day-old fetuses was 48.86±3.13% in the humerus, 53.19±2.37% in the radius, 55.73±4.22% in the ulna, 33.63±2.06% in the femur, 43.20±2.78% in the tibia, and % in the fibula. 
 Conclusion: We think that these results will shed light on the studies to be conducted on the detection of skeletal anomalies in teratological studies and contribute to a more comprehensive evaluation of the findings to be obtained from the studies to be conducted.
