Determination of Hepatotoxic Indospicine in Australian Camel Meat by Ultra-Performance Liquid Chromatography–Tandem Mass Spectrometry

Abstract: Indospicine is a hepatotoxic amino acid found in Indigofera plant spp. and is unusual in that it is not incorporated into protein but accumulates as the free amino acid in the tissues (including muscle) of animals consuming these plants. Dogs are particularly sensitive to indospicine, and secondary poisoning of dogs has occurred from the ingestion of indospicine-contaminated horse meat and more recently camel meat. In central Australia, feral camels are known to consume native Indigofera species, but the preva… Show more

“…The reaction mixture was gradually warmed up to room temperature and stirred for 2 h. Upon completion, saturated aqueous NH 4 Cl solution (15 mL) was added and the aqueous layer was extracted with CH 2 Cl 2 (10 mL × 3). 1 L-2,6-Diamino-6-iminohexanoic acid (9). Purification by flash column chromatography on silica gel ( petroleum ether/ ethyl acetate, 2 : 1 to 1 : 1) gave the desired product as a colourless oil (2.82 g, 95%).…”

“…9 In the reported analysis of residues in camel meat tissues indospicine (1) Finally, in order to test the utility of this synthetic methodology, an indospicine analogue, L-norindospicine (9) was also synthesised from commercially available N-cbz-L-serine ethyl ester (10) (Scheme 3). In order to explore the utility of this route and to produce labelled material for quantitation studies, 9 a multiply deuterated indospicine, [ 2 H 3 ]-1 (Scheme 2) was synthesised employing the same This deuterated indospicine has been demonstrated to be useful as an internal standard in LC-MS/MS stable isotope dilution analysis (SIDA) of indospicine in complex tissue matrixes to compensate for matrix effects and increase accuracy of analysis.…”

“…Van gossypol (katoenplant) is bekend dat het in lage hoeveelheden overgedragen kan worden naar plasma en melk (Wang et al, 2012). Overdracht van indospicine (Indigofera plant)(aminozuur, arginine analoog) naar vlees is gemeld in Australië (FitzGerald et al, 2011;Tan et al, 2014). Het is onbekend of deze planttoxine ook in Nederlandse weideplanten voorkomt of dat geïmporteerd vlees deze planttoxine bevat.…”
