Determination of human alcohol dehydrogenase and acetaldehyde dehydrogenase genotypes by single strand conformation polymorphism in discontinuous buffer electrophoresis

Abstract: Under appropriate conditions single strand conformation polymorphism (SSCP) analysis of polymerase chain reaction (PCR) products allows the detection of single base mutations in a given DNA fragment. We adapted this method for the routine determination of allele variants of human alcohol and acetaldehyde dehydrogenase without radioisotopic labeling. After PCR amplification of the selected exon, the DNA fragments were heat-denatured and loaded on a polyacrylamide gel containing glycerol. For electrophoresis a d… Show more

“…The genotyping of the ADH2, ADH3, and ALDH2 have been described in detail el~ewhere. '~ Briefly, we followed the method of Xu et aL3* for the genotyping of ADH2, the method of Walzer et al 33 for the genotyping of ADH3, and the method of Tu and for the genotyping of ALDH2. All polymerase chain reactions were conducted in a GeneAmp PCR System 9600.…”

“…The method of SSCP/HD analysis used in this study has been successful in identifying polymorphisms in ADH2 and ADH3 within the South African context. A similar SSCP method has previously been reported for genotyping ADH2 (8). A similar SSCP method has previously been reported for genotyping ADH2 (8).…”
