Determination of IL‐1 and IL‐6 Levels in Human Embryo Culture‐Conditioned Media

Abstract: We found an average IL-1 beta level +/- SEM of 82 +/- 4 pg/ml (n = 11) in HECCM from viable pregnancies and a significantly lower value, 14 +/- 2 pg/ ml (n = 25, P < 0.001), in HECCM from embryos that did not lead to viable pregnancies. In control medial the average IL-1 beta level was 11 +/- 1 pg/ml (n = 17), (P < 0.001 vs. HECCM from viable pregnancies). In contrast IL-6 levels were undetectable in all samples analyzed. Judging by our results we suggest that measurement of IL-1 level in HECCM could be a usef… Show more

“…Results in this study indicate that CM prepared with bovine cumulus cells, either serum-containing CM, or BSA-containing CM, yielded similar morula and blastocyst development compared with co-culture with bovine cumulus cells (Table 3). There have, however, been numerous studies reporting the incubation of in vitro produced (IVP) or in vivo embryos in medium conditioned with different cells (Kobayashi et al 1992, 1997, Baranao et al 1997, Liu et al 1998, Joo et al 2001. Cell numbers of blastocysts derived in CM were not different from coculture groups.…”

“…Embryos have also been reported to secrete substances into their surrounding environment (Van Langendoncket et al 1996, Izquierdo et al 1999. Cell-free conditioned medium (CM) has been used in several laboratories in the culture of in vivo and in vitro fertilized embryos (Baranao et al 1997, Funston et al 1997, Ishiwata et al 2000, Galli et al 2003. Studies have shown that CM has similar effects on embryo development to those produced in various co-culture systems.…”

“…Preimplantation embryos are known to produce a number of cytokines including interleukin-1a (Sheth et al, 1991), interleukin-1b (Takacs and Kauma, 1996;Krussel et al, 1998) and interleukin-6 (Zolti et al, 1991;Gerwin et al, 1995). In fact, the production of interleukin-1 had been suggested to be a marker of embryo quality in clinical assisted reproduction (Baranao et al, 1997). Functional response elements of interleukin-1 and interleukin-6 are present in the mouse C3 gene (Kawamura et al, 1992).…”

“…The embryo produces several factors during its development to signal its presence to the maternal organism, and several embryonic factors that modulate endometrial receptivity have been reportedly detected in embryo culture supernatant (ECS) (2)(3)(4)(5)(6)(7)(8). In blastocyst transfer (BT), cross-talk is absent until the blastocysts are transferred and the pregnancy rate (PR) using BT is limited to around 40% (9).…”
