Determination of methadone in human hair by radioimmunoassay

Abstract: The concentrations of methadone in human hair were measured. The washed hair was cut in 1-mm pieces approximately, then incubated overnight at 45 degrees C with 0.1 m HCl. The extracts were alcalized by 1 m NaOH and diluted by phosphate buffer, pH 7.4. The methadone concentrations were determined by radioimmunoassay. The method is simple, rapid, and practicable for routine determination.

“…Radioimmunoassay (RIA) was initially employed for screening hair samples for drugs of abuse, 68,8991 but the majority of laboratories have now replaced RIA with enzyme-linked immunosorbent assays (ELISA). 92 Immunoassays originally developed for whole blood have been adapted or optimized specifically for hair.…”

“…The presence of methadone in human hair was demonstrated by radioimmunologic method (Balabanova and Wolf 1988) but not by GC/MS. This study described the determination of methadone alone in human hair by GC/MS.…”

“…Recently, we reported also the presence of methadon in human hair obtained from patients receiving daily methadone (Balabanova and Wolf 1988). However, data on the occurence of the drug in axillary and pubic hair are not available.…”
