Determination of mite Allergens in House Dust Using the Enzyme Immunoassay

Abstract: The aim of this study was to determine the level of two major mite allergens Dermatophagoides pteronyssinus (Der p 1) and Dermatophagoides farinae (Der f 1) in 30 urban homes in Zagreb, Croatia, using the enzyme immunoassay with two monoclonal antibodies which has been established as the reference method for indoor allergen analysis. Dust samples were taken by vacuuming a carpeted area and collected on cellulose filters. The ranges of Der p 1 and Der f 1 were (0.1-12.5) microg g-1 (median 0.32 microg g-1) and … Show more

“…14 Moreover, recent evidence has shown that removing mite allergen from indoor environments may be of clinical benefit. 52 Thus, we analyzed the Der p 1 concentration in samples obtained at T 1 and T 2 . In each classroom, no significant difference in allergen level was observed ( Table 3, Supporting Information Figure S1).…”

“…The centrifuge tubes were shaken for 2 h at room temperature and centrifuged for 10 min at 3000 rpm (Prester et al, 2007). Then, the supernatants were transferred into Eppendorf tube and stored at − 20°C until analysis of dust mites.…”

“…Concentrations of dust mite allergen Der p 1 (major Dermatophagoides pteronyssinus allergen) were measured by double monoclonal antibody (mAb)-based enzyme immunoassay (ELISA) as described previously (Prester et al 2007). Set of monoclonal antibodies (5H8 and 4C1) and standard of Der p 1 were purchased from Indoor Biotechnologies Ltd (Cardiff, UK).…”
