Determination of Ochratoxin A in Italian Red Wines by Molecularly Imprinted Solid Phase Extraction and HPLC Analysis

Abstract: An extraction method based on molecularly imprinted polymer prepared through a mimic template approach was used for the determination of ochratoxin A in 17 red wines from different geographical regions of Italy. Sample loading (wine sample diluted 1:1 with 1% v/v aqueous solution of PEG 8000), washing (2 mL water/acetonitrile 4:1 v/v), and elution (2 mL of acetonitrile/acetic acid 98:2 v/v) conditions allowed the optimization of the extraction method, capable of preconcentrating ochratoxin A below the maximum … Show more

“…However, the reported LOD and LOQ values (0.025 and 0.08 μg/L, respectively) are about 1.5-fold higher than those obtained in this work. LOD (0.075 μg/L) and LOQ (0.225 μg/L) values reported in a similar study that used another synthesized MIP to determine OTA in Italian red wines were 4.4-fold higher than those of the BSA-based SPE method (Giovannoli, Passini, Di Nardo, Anfossi, & Baggiani, 2014). Additionally, Rhouati, Paniel, Meraihi, and Marty (2011) developed and tested an oligosorbent (aptamer) for the analytical determination of OTA in beer, reporting a LOD of 0.2 μg/L, which is also far over the response obtained herein using albumin as sorbent (0.017 μg L −1 ).…”

“…Thus, the obtained MIPs are able to specifically rebind with the templates in a strong and selective way. Owing to their mechanical and chemical stability, low cost and high selectivity, MIPs have been exploited for catalysis, sensors, drug delivery, solid‐phase extraction (SPE) and chromatographic separation (Wang et al ., ; Resmini, ; Yang and Zhao, ; Bereli et al ., ; Li et al ., , ; Giovannoli et al ., ). Molecularly imprinted solid‐phase extraction (MISPE) is one of the most successful and useful applications for the separation and enrichment of many analytes from a complex matrix (Zeng et al ., ; Scorrano et al ., ; Sadeghi and Jahani, ; Guan et al ., ).…”
