Determination of Optimal Electrospray Parameters for Lipidomics in Infrared-Matrix-Assisted Laser Desorption Electrospray Ionization Mass Spectrometry Imaging

Abstract: Infrared matrix-assisted laser desorption ionization (IR-MALDESI) is an ambient mass spectrometry imaging (MSI) technique that relies on electrospray ionization (ESI) for ion generation of desorbed neutrals. Although many mechanisms in IR-MALDESI have been studied in depth, there has not yet been a comprehensive study of how the ESI parameters change the profiles of tissue specific lipids. Acetonitrile (ACN)/water and methanol (MeOH)/water solvent systems and compositions were varied across a series of applied… Show more

“…Of note, various ammonium adducts were observed, particularly for neutral lipids, although a source of ammonium was not added. Many of these ammonium adducts have been previously reported, including those of triglycerides, using the same or similar ionization methods ( 47 , 48 , 49 , 50 ). Two possible theories for these adducts are given by Zhou et al.…”

“…An ice layer was uniformly deposited on the sample surface as a mid‐IR absorbing matrix by cooling the sample stage down to −8°C and was maintained throughout the experiment by keeping the relative humidity inside the sample enclosure at 10 ± 2% 31 . The electrospray solvent of 50:50 v/v acetonitrile/water with 1 mM formic acid 32 was injected at 1.0 μL·min −1 flow rate to ionize the ablated neutral materials via an electrospray and transmit them into the mass spectrometer.…”

“…To determine the optimal sample composition, 100-μM caffeine, and vent compositions were previously found to be optimal for lipidomic IR-MALDESI-MSI analyses. 30 The abundances of each lipid were summed together and then averaged with the average caffeine abundance as a measure of target ion abundances over a wide m/z range.…”
