Determination of some anti-human immunodeficiency virus nucleosides by capillary zone electrophoresis-tandem mass spectrometry

Abstract: We describe the analysis of two potent anti-human immunodeficiency virus reverse transcriptase (RT-HIV) inhibitors, zidovudine (AZT) and stavudine (d4T), among a pool of natural nucleosides (A, C, G, U, T) by capillary zone electrophoresis (CZE)-ionspray-tandem mass spectrometry (MS/MS) in positive ion mode. Several volatile formic acid-ammonia buffers having the same ionic strength (50 mM) but different pH values varying in the 9-11 pH range were prepared and tested to determine the best electrophoretic migra… Show more

“…[19][20][21][22] Hyphenated CE-MS systems were also used. 23 A few CE methods were reported for the determination and validation of antiretrovirals drugs in bulk drugs and pharmaceutical formulations. 24,25 Sekar and Azhaguvel, 25 separated and determined lamivudine and stavudine with the nevirapine (NNRTI) in pharmaceutical formulations by micellar electrokinetic capillary chromatographic (MEKC) method, all antiretroviral drugs were separated within 14 min using 10 mmol L -1 sodium tetraborate (pH 9.8), 100 mmol L -1 SDS and 15% 2-propanol as an organic modifier.…”

“…Additionally, MS detection seems to be recommendable for the determination of low concentrations of some PIs as they may present poor absorption in the UV range (e.g., lopinavir) [6,12,[20][21][22]. Apart from HPLC, capillary electrophoresis (CE) has also been used for the determination of drug mixtures in various biological matrices [23][24][25]. It should be remarked that the principal shortcomings of CE methods are the poor detection limits, ranging from 0.2 to 1.0 g mL −1 .…”
