SUMMARYPreservation of epididymal spermatozoa is important to save genetic material of endangered species and breeds, or in case of unexpected injury, which will end the breeding career of valuable sires. Seminal plasma (SP) influences sperm quality in a dosedependent manner and its addition to preserved semen immediately before insemination may be beneficial for sperm fertility. Increased plasma membrane stability of epididymal spermatozoa reduces freezing injury of cells, and the addition of SP after freezing and thawing might have activating and protecting effects on spermatozoa within the female genital tract. In this study, epididymal spermatozoa were harvested by retrograde flush of the epididymal cauda immediately after routine castration and frozen. Seminal plasma was collected from other six stallions. Homologous SP (SP from the same species, but from a different animal) was added to frozen-thawed epididymal spermatozoa at concentrations of 0, 5, 20, 50 and 80% SP. Addition of SP increased sperm motility and influenced kinematic values in a dose-dependent manner (p < 0.05). Motility improved at concentrations of 20 and 50% SP, but did not further increase at 80% SP. There was no difference in sperm motility among SP from six different donor stallions regardless of the concentrations of SP (p > 0.05). Total and progressive motility of ten frozen-thawed epididymal spermatozoa samples collected from different stallions after dilution with extender and 5, 20, 50 or 80% SP differed significantly (p < 0.05). In conclusion, addition of homologous SP to frozen-thawed stallion epididymal spermatozoa immediately improved motility in a dose-dependent manner regardless of semen quality of SP donor stallions. This might positively influence fertility when SP is added before insemination. Moreover, there seems to be a threshold level of SP concentration for optimal improvement of sperm motility.

In order to obtain a successful outcome in artificial insemination programs, a few precautions must be taken with regard to male breeders. It is established that environmental factors can change the hormonal secretion, the cell differentiation that occurs in the testicles, as well as sperm maturation and transport in the epididymis. In view of adverse nutritional factors, reproductive organs can present degeneration and disorders in different degrees and intensities, affecting the animal fertility. The term nutraceutical describes products derived from foods, which may provide additional health benefits, beyond the basic value found in diets. Among the main nutraceuticals used in male reproduction, there are omega-3, arginine, B-complex vitamins, L-carnitine, β-carotene and antioxidants. The aim of this paper is to present the main substances used as nutraceuticals, in order to improve semen quality, obtaining most favorable outcomes of stallions in the use of natural breeding or artificial insemination.Keywords: artificial insemination, fertility, nutrition, semen, spermatogenesis.resumo Para um sucesso em programas de inseminação artificial, algumas precauções devem ser tomadas em relação aos reprodutores machos. Um déficit nutricional pode levar a degeneração e distúrbios dos órgãos reprodutivos em diferentes graus e intensidade, afetando a fertilidade. O termo nutracêutico descreve produtos derivados dos alimentos, que podem fornecer benefícios adicionais a saúde, além dos valores básicos encontrados na dieta. Entre os vários nutracêuticos utilizados na reprodução, temos o ômega-3, arginina, vitaminas do complexo B, L-carnitina, β-caroteno e antioxidantes. Objetivou-se com essa revisão apresentar as principais substâncias utilizadas como nutracêuticos, com a finalidade de melhorar a qualidade seminal, obtendo resultados favoráveis dos garanhões com a monta natural ou a inseminação artificial.Palavras-chave: inseminação artificial, fertilidade, nutrição, sêmen, espermatogênese.

Um método simples e rápido para a determinação de Ca, Cu, Fe, Mg, Mn, Se e Zn em sêmen bovino por espectrometria de massas com plasma indutivamente acoplado (q-ICP-MS) é descrito. Previamente as análises, 200 µL de amostras foram diluídas 1:50 em solução contendo Triton ® X-100 (0,01% v/v) e ácido nítrico (0,5% v/v). Os limites de detecção foram de 0,3, 0,03, 0,2, 0,04, 0,04, 0,03 e 0,03 µg L -1 para 44 Ca, 63 Cu, 57 Fe, 24 Mg, 64 Zn, 82 Se e 55 Mn, respectivamente. Para efeitos de comparação e validação do método, quatro amostras de sêmen bovino foram analisadas por ICP-MS pelo método proposto e por espectrometria de absorção atômica com chama (FAAS) ou espectrometria de absorção atômica em forno de grafite (GF AAS), e não foram encontradas diferenças estatísticas entre as técnicas com aplicação do teste-t (95% de confiança). Então, o método proposto foi aplicado na determinação de Ca, Cu, Fe, Mg, Mn, Se e Zn em amostras de sêmen bovino coletadas de diferentes raças, as quais são usadas em programas de reprodução animal e inseminação artificial.A simple and fast method for the determination of Ca, Cu, Fe, Mg, Mn, Se and Zn in bovine semen by quadrupole inductively coupled plasma spectrometry (q-ICP-MS) is described. Prior to analysis, samples (200 µL) were diluted 1:50 in a solution containing 0.01% v/v Triton ® X-100 and 0.5% v/v nitric acid and directly analyzed by ICP-MS. The limits of detection of the method are 0.3, 0.03, 0.2, 0.04, 0.04, 0.03 and 0.03 µg L -1 for 44 Ca, 63 Cu, 57 Fe, 24 Mg, 64 Zn, 82 Se and 55 Mn, respectively. For purposes of comparison and method validation, four ordinary bovine semen samples were directly analyzed by ICP-MS and by flame atomic absorption spectrometry (FAAS) or graphite furnace atomic absorption spectrometry (GF AAS), with no statistical difference between the techniques at the 95% level when applying the t-test. Then, the proposed method was applied in the determinations of Ca, Cu, Fe, Mg, Mn, Se and Zn in collected samples of bovine semen from different breeds, which are used in reproduction programs and artificial insemination.
