Determination of T-2 and HT-2 Toxins in Cereals Including Oats after Immunoaffinity Cleanup by Liquid Chromatography and Fluorescence Detection

Abstract: A reliable method for the determination of T-2 toxin and HT-2 toxin in different cereals, including oats, as well as in cereal products was developed. After extraction with methanol/water (90/10, v/v) and dilution with a 4% NaCl solution, the toxins were purified with immunoaffinity columns, derivatized with 1-anthroylnitrile, separated by HPLC, and determined using fluorescence detection. Due to the unspecific derivatization reagents, validation parameters were matrix dependent: in the range 10-200 microg/kg,… Show more

“…Production of mycotoxins and analyses DON was produced by Fusarium graminearum (NRRL 5883) and T-2 toxin by Fusarium sporotrichioides (NRRL 3299) strains on corn substrate according to Fodor et al (2006). DON and 15-acetyl DON content of the feed was determined according to Pussemier et al (2006), while T-2 and HT-2 concentration was measured by HPLC after immunoaffinity cleanup (Trebstein et al, 2008).…”

“…1-antroylnitrile (1-AN) was provided by Wako (SOBIODA SAS, France). DMAP and 1-AN were prepared in toluene as reported by Trebstein, Seefeldet (16). The HPLC-FLD methodology used in this study was adapted from Lippolis, Pascale (17), Pascale, Haidukowski (18), Trebstein, Seefeldet (16), Trebstein,Marschik (19), and Visconti, Lattanzio (20) to measure the T-2 toxin in liquid cultures.…”
