Determination of the Therapeutic Mean Effective Doses of Several Anti-inflammatory Agents in Adjuvant Arthritic Rats

Abstract: The large variation in the severity of the arthritic response of the adjuvant-injected rat often makes it impossible to obtain statistically manageable dose–response curves with anti-inflammatory drugs. Consequently, the relative potency of anti-inflammatory drugs generally was not established. In the present study, with a modification of the therapeutic test, reliable dose–response curves were obtained with seven anti-inflammatory drugs. With this method the "therapeutic" mean effective dose (ED50) and relati… Show more

“…On the 9th day after CFA injection the animals for the final experiment were selected. Criterion for the selection was the volume of the right hind paw between 14 and 16 mm Hg [4]. Rats were then randomized and divided into five groups (n ¼ 9 per group) and received p.o.…”

“…Etodolac is structurally related to prodolic acid, and like this compound, which for a while was considered for clinical trials (92), it is more active against the chronic than against the acute model of inflammation (93). The product is a good analgesic and its therapeutic index is 3 and 4 times higher than that of PB and indomethacin (94).…”

“…Male Sprague-Dawley rats, each weighing about 250 g, were injected intradermally into the plantar region of the right hindfoot with 0.05 ml of a fine suspension of killed and dried Mycobacterium butyricum (Aoyama B strain) at a concentration of 5 mg/ml in liquid paraffin (13). The volumes of both paws were measured daily for 14 days using a mercury plethysmograph.…”
