Determination of Tolterodine tartrate in bulk and formulation by extractive colorimetric method using Tropaeolin OOO-1

Schinopsis brasiliensis Engl. is a native plant of Caatinga which has high concentrations of compounds capable of absorbing ultraviolet light, suggesting its potential application for the development of sunscreen preparations. After its identification and collection, this vegetable drug was submitted to a physicochemical analysis through the preparation of ethanolic extract. The phytochemical screening and analysis of extracts were carried out by high-performance liquid chromatography (HPLC) evaluation. The antioxidant activity of the extract was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) method and β-carotene bleaching test. Inhibitory hemolytic activity and morphological deformation of erythrocytes induced by H2O2 were also demonstrated and the antimicrobial activity was analyzed by the minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) method. For the in vitro determination of the sun protection factor (SPF), the spectrophotometric method was used. From the analyses carried out with this species, this plant showed significant results for the antioxidant and antimicrobial activities, as well as sunscreen action. Important flavonoids were identified. These data are an important step for the development of new photoprotective cosmetic with Caatinga species, revealing importance and representing another incentive for the preservation of the species involved and analyzed in the study.

A rapid, sensitive and accurate ultra-performance reversed phase liquid chromatographic method was developed for the simultaneous determination of tamsulosin and tolterodine in pure form and pharmaceutical preparation. The developed UPLC method is superior to conventional HPLC with respect to speed, resolution, solvent consumption and cost. The separation was carried out on RP C 18 nucleosil (1.7 µm, 5 cm x 2 mm) using an isocratic mode in eluting Tamsulosin and Tolterodine at 1.54 min and 2.43 min respectively with a mobile phase composed of acetonitrile and 0.025N potassium phosphate buffer pH 3.50 (60%:40%), respectively. Chromatographic run time was 5 min with a flow rate 0.5 ml/min and UV detection at 220 nm. The linearity for tamsulosin and tolterodine were in the range of 2-20 µg/mL for both drugs, showed excellent recoveries for bulk and tablet dosage form with a very low LOD of 4.29 and 0.59 ng/mL for tamsulosin and tolterodine, respectively. The method has been validated for linearity, accuracy, precision, specificity, and limit of detection, limit of Quantification, robustness, and ruggedness. The method which was developed was validated as per the ICH guidelines. Finally, the method was compared statistically with reference methods indicating that there is no significant difference between them in respect of precision and accuracy.

Objective: Simple, sensitive, and accurate spectrophotometric methods have been developed for the assay of tolterodine tartrate (TOL) in bulk drug and pharmaceutical formulations. Methods:The proposed methods are based on oxidation reaction of TOL with a known excess of cerium(IV) ammonium sulfate as an oxidizing agent in acid medium followed by determination of unreacted oxidant by adding a fixed amount of dye, e.g., amaranth (AM), rhodamine 6G (Rh6G), and indigo carmine (IC) followed by measuring the absorbance at 520, 530, and 610 nm, respectively. The effect of experimental conditions was studied and optimized.Results: The Beer's law was obeyed in the concentration ranges of 1.0-10, 1.0-12, and 0.5-9.0 μg/mL using AM, Rh6G, and IC dyes, respectively, with a correlation coefficient ≥0.9995. The calculated molar absorptivity values are 1.868×10 4 , 1.008×10 4 , and 1.623×104 L/mol/cm using AM, Rh6G, and IC dyes, respectively. The limits of detection and quantification were reported. Intraday and interday accuracy and precision of the methods have been evaluated. No interference was observed from the additives. Conclusion:The proposed methods were successfully applied to the assay of TOL in tablets preparations, and the results were statistically compared with those of the reported method by applying Student's t-test and F-test. The reliability of the methods was further ascertained by performing recovery studies using the standard addition method.
