A comparative study of the primary and secondary metabolites of the juice of several mandarin cultivars with different pollination, seed production and parthenocarpic abilities was carried out, and the antioxidant capacity of the hydrophilic fraction was measured. Correlation by a Principal Component Analysis and a Cluster Analysis was used. By a multivariate analysis, the 15 citrus cultivars were clustered into four groups consistently with citrus types. The presented data are an important factor for choosing varieties with high potential as a nutraceutical source. These aspects are necessary as consumers demand prevention of health problems through nutrition and certain fruit quality traits, including fruit size, internal quality, good rind color and easy peeling.

Volatile organic compounds (VOCs) in plants are involved in aroma and pest resistance. These compounds form a complex mixture whose composition is specific to species and often to varieties. Despite their importance as essential factors that determine peach fruit quality, understanding of molecular, genetic, and physiological mechanisms underlying aroma formation is limited. The aim of this study was the identification in peach of quantitative trait loci (QTLs) for fruit VOCs to understand their genetic basis using an F1 population of 126 seedlings deriving from the cross between "Bolero" (B) and "OroA" (O), two peach cultivars differing in their aroma profile. Dense single nucleotide polymorphism (SNP) and SSR maps covering the eight linkage groups of the peach genome were constructed by genotyping with the International Peach SNP Consortium peach SNP array v1, and data for 23 VOCs with high or unknown "odor activity value" were obtained by gas chromatography-mass spectrometry analysis of fruit essential oil in the years 2007 & Genomes (2013) 9:189-204 DOI 10.1007 QTLs were identified, most consistent in both years. QTLs were identified for the 23 VOCs studied, including three major QTLs for nonanal, linalool, and for p-menth-1-en-9-al stable in both years. Collocations between candidate genes and major QTLs were identified taking advantage of the peach genome sequence: genes encoding two putative terpene synthases and one lipoxygenase (Lox) might be involved in the biosynthesis of linalool and p-menth-1-en-9-al, and nonanal, respectively. Implications for marker-assisted selection and future research on the subject are discussed.

Method for the Determination of Benzene Metabolite t,t-Muconic Acid in Urine by HPLC-UV with an Ion Exclusion Column

Abstract:The benzene metabolite trans,trans-muconic acid (tt-MA) is widely used as a biological indicator of exposure to this xenobiotic. An analytical method was developed and validated for the determination of urinary tt-MA using solid phase extraction onto a Strata SAX cartridge. The analysis was performed by HPLC with an Aminex HPX-87H ion exclusion column and a UV diode array detector. The sample preparation conditions were optimized using a 2 4´1 factorial design.Comparison of the slopes of standard analytical curves prepared in aqueous solution and urine showed that the biological matrix suppressed the tt-MA signal by around 50%, so the analytical curves were prepared with tt-MA standard at low concentrations in pooled urine. The analytical curves in the range of 5-500 µg¨L´1 showed determination coefficients values (R 2 ) > 0.99 for tt-MA standard in water and R 2 > 0.98 for tt-MA standards in pooled urine. The coefficients of variation obtained using seven replicates were lower than 3.6%, and recoveries of tt-MA from solutions containing 5, 25, 50 and 100 µg¨L´1 of the analyte were in the range 85%-90%, demonstrating the satisfactory precision and accuracy of the method. The limits of detection and quantification were 0.11 and 0.36 µg¨L´1, respectively. The benefits of this new method developed is the possibility of complete chromatographic peak separation for the determination of tt-MA at baseline, without matrix components' interference as normally found in the C18 column. This is the first time that this chromatographic column has been used for the analysis of tt-MA in urine.
