Developing a System for Regulating Expression of Human Hepatocyte Growth Factor Using Tetracycline in NRK52E Cells

Abstract: Introduction: Hepatocyte growth factor (HGF) is a target of gene therapy for renal fibrosis. The aim of this study was to establish a human HGF gene expression system that is regulated by tetracycline (Tet) in normal rat kidney tubular epithelial cells (NRK52E cells). Materials and Methods: The plasmids pTet-on, pBI-L-HGF and pTK-Hyg were transfected sequentially into NRK52E cells using Lipofectamine 2000. The expression of HGF gene was measured, and the activity of expressed HGF was detected. Results: A clone… Show more

“…On the bais of these results, we suggested 2 phases in this iron-induced cell damage model: an injury phase (3 hours) and a recovery proliferative phase (24 hours). Considering that HGF is not expressed in renal proximal tubular cells, 25 we evaluated the expression of its F I G U R E 5 Effect of anti-miR-26a on cells exposed to Fe 3+ . (A) Anti-miR was efficient to significantly suppress miR-26a expression;…”

“…On the bais of these results, we suggested 2 phases in this iron‐induced cell damage model: an injury phase (3 hours) and a recovery proliferative phase (24 hours). Considering that HGF is not expressed in renal proximal tubular cells, we evaluated the expression of its receptor, c‐Met, and STAT3, a molecule downstream of HGF action. There was an increase in c‐Met gene expression at 3 hours (damage phase), suggesting that the cells, in spite of the damage, may be prone to respond to HGF.…”
