Development and biological activity of long-acting recombinant human interferon-α2b

Abstract: Background: The type I human interferon (IFN) family consists of a group of cytokines with a multiplicity of biological activities, including antiviral, antitumor, and immunomodulatory effects. However, because the half-life of IFN is short, its clinical application is limited. Increasing the yield and biological activity of IFN while extending its half-life is currently the focus of IFN research. Results: Two novel long-acting recombinant human IFN-α2b (rhIFN-α2b) proteins were designed in which the carboxyl-t… Show more

“…The interferon alpha (IFNα) belongs to the I type of interferons, which defines this molecule as being possible to be formed by each nucleated cell and having immunomodulatory and antineoplastic properties. 61 Due to its limiting effect on fibroblast proliferation and modulation of collagenase and metalloproteinase levels, IFN-α-2b has been proposed as a possible treatment for PD. 62 Historically, effectiveness has been shown in a single-blind, placebocontrolled study.…”

“…The endogenous interferons are generated by the cells in response to infections due to their antiviral features. The interferon alpha (IFN‐α) belongs to the I type of interferons, which defines this molecule as being possible to be formed by each nucleated cell and having immunomodulatory and antineoplastic properties 61 . Due to its limiting effect on fibroblast proliferation and modulation of collagenase and metalloproteinase levels, IFN‐α‐2b has been proposed as a possible treatment for PD 62 …”

“…Fusion protein technology is widely used to prolong the stability of proteins. Carboxyl-terminal peptide (CTP) fusion is one such strategy (Zhang et al 2020). CTP is the β-subunit of human chorionic gonadotropin, and comprises 28 amino acids and 4 O-linked oligosaccharides (Birken and Can eld 1977).…”

“…15 The IFN-α2b also can be used to treat the human immunodeficiency virus (HIV) and various cancers, such as: malignant melanoma, chronic myeloid leukemia and renal cell carcinoma. 17 Many E. coli strains are employed for recombinant DNA transformation protein productions. These strains include DH5α competent cells for cloning 18 and the BL21(DE3) competent cell for protein expression.…”
