Development and characterization of five rainbow trout pituitary single-cell clone lines capable of producing pituitary hormones

Abstract: Five single-cell clone lines (mRTP1B, mRTP1E, mRTP1F, mRTP1K, and mRTP2A) have been developed from adult rainbow trout pituitary glands. These cell lines have been maintained in a CO 2 -independent medium supplemented with 10% fetal bovine serum (FBS) for more than 150 passages. At about 150 passages, the doubling time of each single-cell clone in a CO 2 -independent medium supplemented with 10% FBS at 20 8C was 3 . 6G0 . 7, 2 . 8G0 . 7, 3 . 2G0 . 8, 5 . 5G0 . 6, and 6 . 6G0 . 6 days respectively. Each singl… Show more

“…This is likely due to the increased in concentration of dose which had the satisfactory action on the female's gonads. According to Chen et al [3], the synchronization of ovulation in fish after the application of pituitary dose is very important aspect. Egg weight, egg number and percentage egg weight were higher in 4 mg/kg dosage than the other dosages.…”

“…Demand for fish in both rural and urban areas is very high and continues to grow in Tanzania by FAO in 2007 [1]. The African catfish (Clarias gariepinus) has gained popularity in the aquaculture sector of Tanzania in the recent times [2,3]. Its demand has increased as food, control of over-population in tilapia ponds and as bait for the Nile perch fishery [4][5][6][7].…”

“…Pituitary tissue culture requires the pituitary from many individuals for experiment because the fish pituitary is very small. Thus, differences between individuals can lead to a number of variations within or between experiments ( Chen et al, 2010 ). Pituitary cell culture requires the process of separating and culturing pituitary cells, which is difficult to reproduce consistently, and recovery periods can affect specific gene expression ( Chen et al, 2010 ).…”
