Development and comparison of the methods for quantitative electron probe X‐ray microanalysis analysis of thin specimens and their application to biological material

Abstract: In recent years, there has been a return to the use of electron probe X-ray microanalysis for biological studies but this has occurred at a time when the Hall programme which acted as the mainstay for biological microanalysis is no longer easily available. Commercial quantitative routines rely on the Cliff-Lorimer method that was originally developed for materials science applications. Here, the development of these two main routines for obtaining quantitative data from thin specimens is outlined and the limit… Show more

“…cellulose) to the samples measured under the same analytical conditions (Tylko et al, 2010). Although quantification algorithms in thick and thin samples are well defined (Warley, 1997(Warley, , 2016, analysis of semi-thick samples is still limited (Tylko, 2013(Tylko, , 2016.…”

“…Since the liver contains variable amounts of iron and copper in the cytoplasm and organelles, the results of X‐ray spot microanalysis and 2‐dimensional X‐ray imaging should be evaluated differently . The element content is expressed by the ratio of the molar concentration or weight to the total elements in the spot tissue analyzed, while 2‐dimensional images of each element can be obtained when an organelle contains more of an element than the adjacent cytoplasm.…”
