Development and validation of a hydrophilic interaction liquid chromatography–tandem mass spectrometry method for determination of isoniazid in human plasma

Abstract: An LC-MS/MS method for the determination of isoniazid in human plasma was developed and validated. Human plasma aliquots of 100 μL were used for analysis. The assay used nialamide as the internal standard. The calibration curve concentration range was 50 to 10,000 ng/mL. Sample preparation utilized protein precipitation, and the supernatant was directly injected onto silica column without reconstitution. The recovery was over 90% and matrix effect was negligible. The method is simple and fast, which is advanta… Show more

“…Bench-top stability testing and the more extensive stability assessment showed that isoniazid is unstable in human serum unless it is freeze-dried or stored at -80°C. Instability of isoniazid in plasma at ambient temperature and -20ºC has been reported earlier [28,29]. This instability calls for adequate standard operating procedures and rapid handling of samples.…”

“…Therefore, an experiment in fivefold was performed to evaluate the magnitude of the carry-over by injecting five LLOQ (Lower Limit of Quantification) samples after a HLQ [28,29]. The total amount of stock solutions added to the serum was less than 5% of the final volume.…”

“…HPLC-UV [7][8][9][10][11] and HPLC-MS [12,13] methods using RP sorbents dominate the simultaneous analysis of INH and AcINH. Hydrophilic interaction liquid chromatography (HILIC) on Hypersil silica with MS detection has been also developed for determination of INH in plasma [14]. Vitamin B6 is typically analyzed in RP settings after pre or post column derivatization using fluorescent [15][16][17][18][19] or electrochemical detections [20].…”

“…It then had to be injected two times for each sample to analyze PZA, INH and RFP. LC-MS is selective and sensitive, and several HPLC-MS methods 17,[19][20][21][22][23] have been reported. However, it is very expensive and not always available, particularly in remote areas of developing countries.…”
