Development and Validation of a High-Performance Liquid Chromatography Assay for Voriconazole

Abstract: An analytical method for the determination of voriconazole (UK-109,496; Pfizer) in plasma was developed and validated. The method utilizes solid-phase extraction technology and high-performance liquid chromatography. The lower limit of quantitation is 0.2 g/ml, and the range of linearity tested was 0.2 to 10 g/ml.Voriconazole (VRC; 496 [C 16 H 14 N 5 OF 3 ]; Pfizer Pharmaceuticals) is a novel broad-spectrum triazole antifungal that is used in the treatment of a wide range of opportunistic fungal infections, i… Show more

“…Steady-state trough blood levels of voriconazole were drawn before the morning dose and measured at least 5 days after the drug was initiated and measured as previously described. 11,12 Although the period following stem cell transplantation includes the administration of many drugs metabolized by the cytochrome P450 system, there were no drugs given concomitantly that were known to change voriconazole disposition.…”

“…Steady-state plasma voriconazole trough levels were monitored using a high-performance liquid chromatography (HPLC) assay, 9 5 days to 7 months (median 15 days) after starting the drug or changing the dose. The voriconazole dose was increased if the level was o0.5 mg/ ml and decreased if the level was 47 mg/ml.…”
