Development of a drug resistance model for hepatoblastoma

Abstract: Abstract. Multidrug resistance (MDR) is a major reason for poor treatment results in hepatoblastoma (HB). The objective of this study was to establish a drug resistance model for HB to analyse alternative treatment options in vitro. Both HB cell lines HUH6 and HepT1 were xenotransplanted in NMRI mice (nu/nu) and 2 cycles of cisplatin (CDDP) treatment were administered. Thereafter, xenotransplants were excised and viable tumour cells were re-cultured. 3D cultures of HUH6 and HepT1 cells were generated on a low … Show more

“…Hep3B cells were isolated from a young donor with HCC. The background of a HBV infection in these patients is obvious, as the Hep3B cells expressed constitutive HBsAg [12]. In contrast to Hep3B, the primary tumour of the HC-AFW1 line emerged from a background without any infections, which is the clinical situation in most cases of paediatric HCC in Europe.…”

“…A 1688 bp fragment overlapping the exon 2 to 6 from CTNNB1 was amplified, sequenced and aligned to AY081165 [11]. Same primers were used in the RT-PCR to amplify a 831 bp gene fragment [12].…”

“…Hepatocellular carcinoma (HCC) is one of the most common malignant cancers which is closely associated with chronic infection by the hepatitis B (HBV) and C (HCV) viruses in various populations (30-33). High rate of mortality, cirrhosis risk development, and HCC may occur in the long time unresponsiveness treatment of patients with HCV infection (31, 34-36).…”
