Development of a Monoclonal Antibody-Based Immunochemical Assay for Liquiritin and Its Application to the Quality Control of Licorice Products

Abstract: Liquiritin was reacted with a keyhole limpet hemocyanin (KLH) to synthesize a liquiritin-KLH conjugate as an immunogen for mice. A hybridoma cell line named 2F8 secreted a monoclonal antibody (mAb) against liquiritin, which was applied to an enzyme-linked immunosorbent assay (ELISA) for liquiritin. ELISA showed a good linear range from 0.39 to 25 μg/mL of liquiritin. The maximum relative standard deviation (RSD) values for the intra-assay and interassay were approximately 5%. The recovery rates of liquiritin w… Show more

“…Flavonoids is another another group of bioactive constituents in G. uralensis , of which liquiritin, liquiritgenin, isoliquirtin, isoliquiritigenin, and licochalcone ( Figure 1 ) represent the major ones (Asl and Hosseinzadeh, 2008; Fujii et al, 2014). Effects of AMF on the flavonoids in liquorice under nutrient deficiency have not been thoroughly explored.…”

“…Quality control of licorice root is needed to assure uniformity in the concentrations of these compounds, which can differ according to licorice species, growth place, and harvest season, and so on. To this end, we previously prepared MAbs against GC [4] and Liq [16] and developed an eastern blotting system for detection of GC and Liq and their aglycones (glycyrrhetinic acid and liquiritigenin, respectively) [17] in order to make evident the resource of licorice for further researches. In this method, crude licorice extract and/or traditional Chinese medicine extract are developed on a Thin acrosome reaction in sperm.…”

“…Furthermore, a new staining system, eastern blotting was developed [15] and immunoaffinity isolation of glycyrrhizin resulting in glycyrrhizin-knockout extract, which can be used to survey the real activity of glycyrrhizin in licorice [16,17]. Regarding flavonoid, anti-liquiritin MAb [18] and eastern blotting [19] was developed. Since the above knowledge of components in licorice have been accumulated and also Tanaka et al have been investigating about proteins [20][21][22] and mitochondria [23] related to fertilization using reproducing assay system, we have started to survey in vitro fertilization (IVF) active components in licorice.…”
