Development of a PCR-based mating-type assay for Clavicipitaceae

Abstract: This study developed a PCR‐based mating‐type assay for Clavicipitaceae. PCR primer sets for the mating‐type genes MAT1‐1‐1 and MAT1‐2‐1 were designed based on the amino acid sequences of the conserved alpha and HMG boxes, respectively. The PCR‐based mating‐type assay could be applied for various clavicipitaceous genera (Balansia, Claviceps, Cordyceps and Epichloë). Most of the clavicipitaceous fungi possessed either MAT1‐1‐1 or MAT1‐2‐1, and were supposed to be heterothallic. Although the PCR products obtained… Show more

“…PCR products were purified using a Qiagen QIAquick Gel Extraction Kit and then sent for direct sequencing in both directions using the same primer pair. The 500 bp long ITS sequence obtained (GenBank accession number: DQ374646) was identical with that of P. farinosus strain IFO7866 reported by Yokoyama et al (2004) (GenBank accession number AB083033) and also with some other ITS sequences of various P. farinosus strains available in DNA databases, thus confirming the identity of our isolate MYA-3388.…”

“…In contrast, DNA-based approaches are relatively inexpensive and usually provide reliable mating type assignments (Cherif et al 2006;Dyer et al 2001;Yokoyama et al 2004). These DNA-based methods are, however, dependent on the availability of sequence information for the MAT locus, because MAT idiomorph-specific PCR assays exploit the inherent differences in the MAT genes (Dyer et al 2001;Steenkamp et al 2000).…”

“…The 18S rDNA nucleotide sequence of the isolate 8-2 was identical to that of P. nostocoides JCM 8437, accession number AB104884, in the GenBank database (Yokoyama et al 2004). Therefore, we named this isolate P. nostocoides NTU-FC-LP01 and deposited it in the Bioresource Collection and Research Center (BCRC) in Taiwan, with the accession number BCRC 34144.…”
