Development of an Efficient Method for Recovery of Puumala and Puumala-Related Viruses by Inoculation of Mongolian Gerbils

Abstract: ABSTRACT. Puumala (PUU) virus and PUU-related viruses are difficult to isolate in cell culture. To determine whether animal inoculation would be a better alternative for virus recovery, the Sotkamo strain of PUU virus was inoculated into several animal species. Newborn Mongolian gerbils (MGs), mice, and rats were infected with the Sotkamo strain by intracerebral (ic), intraperitoneal (ip), and subcutaneous (sc) inoculation. Antibodies to PUU appeared in MGs at 30 days post-infection (dpi), and in mice and rats… Show more

“…The Mongolian gerbil is a rodent native to the Mongolian steppe, belonging to the subfamily Gerbillinae ( Zorio et al ., 2019 ). Further, it is an animal used to model infections with various viruses, including Borna disease virus, La Crosse encephalitis virus, encephalomyocarditis virus, Puumala and Puumala-related viruses, and human hepatitis E virus ( Matsuzaki et al ., 1989 ; Osorio et al ., 1996 ; Nakamura et al ., 1999 ; Lokugamage et al ., 2003 ; Hong et al ., 2015 ). EV71 and CVA16 infections in Mongolian gerbils result in disease signs similar to those in humans ( Xu et al ., 2015 ; Sun et al ., 2016 ).…”

“…During the summer of 2003, three gerbils were shown to be infected with monkey pox virus after being exposed to the virus from an infected Gambian giant rat (Kulesh et al, 2004). Newborn gerbils have been shown to be a better model for Puumula virus transmission than rats or mice (Lokugamage et al, 2003). Newborn gerbils can also be experimentally infected with reovirus 3 with histopathologic lesions noted in the brain and pancreas (Yukawa et al, 1993).…”

“…After fixation, the slides with the fixed cells were washed with distilled water, air dried, and stored at Ϫ30 C until use as the antigen slide. The sera from wild rodents were tested for anti-hantavirus antibodies by using the antigen slide and FITC-conjugated protein G (Zymed) (34,39).…”
