Development of HPLC Method for Catechins and Related Compounds Determination and Standardization in Miang (Traditional Lanna Fermented Tea Leaf in Northern Thailand)

Abstract: High performance liquid chromatography (HPLC) for catechins and related compounds in Miang (traditional Lanna fermented tea leaf) was developed to overcome the matrices during the fermentation process. We investigated a variety of columns and elution conditions to determine seven catechins, namely (+)-catechin, (−)-gallocatechin, (−)-epigallocatechin, (−)-epicatechin, (−)-epigallocatechin gallate, (−)-gallocatechin gallate, (−)-epicatechin gallate, as well as gallic acid and caffeine, resulting in the developm… Show more

“…LOD and LOQ were determined as 4 and 9 standard deviations from the blank signal (N = 7), respectively. The results indicated the method's validity [18]. Using the calibration data, the loading percentage of catechin in chitosan was obtained: 40.25%.…”

“…Drug release is the process by which drug solutes travel from their early location in the carrier scheme to the carrier's outer surface and formerly to the release medium [18]. To govern the outline of drug release from catechin-loaded chitosan nanogels, 300 mL phosphate buffer was transferred into 3 beakers.…”

“…This strategic choice aimed to achieve two key objectives: reducing analysis time and enhancing the separation between the seven catechins, as well as gallic acid (GA) and caffeine (Caf). By leveraging these specific components and employing gradient elution techniques, researchers endeavored to optimize the efficiency and accuracy of catechin analysis in tea samples [61].…”

“…Catechins and caffeine were analyzed using a C18 column (250 × 4.6 mm, PhenomenexGemini, Torrance, CA, USA) with the HPLC conditions as follows; UV detector; mobile phase (A) 0.1% acetic acid in acetonitrile and mobile phase (B) 0.1% acetic acid in deionized (DI) water at a flow rate of 1.0 mL/min at 20 °C and injection volume of 10 μL. The concentrations of catechins and caffeine were calculated, based on their RT, compared with the standards that included epigallocatechin gallate (EGCG), epicatechin gallate (ECG), gallocatechin gallate (GCG), epigallocatechin (EGC), epicatechin (EC), catechins (C), gallocatechin (GC) and caffeine (Sigma-Aldrich, St. Louis, MO, USA) [ 25 ].…”
