Development of microsatellite markers for the endangered grassland perennial herb Vincetoxicum atratum (Apocynaceae–Asclepiadoideae)

Abstract: Eight microsatellite markers were developed for the endangered grassland perennial herb Vincetoxicum atratum. The number of alleles ranged from 4 to 14, and the expected heterozygosities were from 0.575 to 0.933 in a population of V. atratum. Five of the eight loci did not significantly deviated from the Hardy-Weinberg equilibrium. All eight loci were tested for cross-species amplification in five other species of Vincetoxicum in Japan. These microsatellite loci will be useful for conservation genetics of V. a… Show more

“…Total DNA was extracted from approximately 100 mg of leaf tissue based on the method of Maki, Horie and Yokoyama [ 38 ]. Ten nuclear simple sequence repeat (nrSSR) loci including vinc5, vinc107, vinc118 [ 39 ], Vpy 012, Vpy 013, Vpy 016, Vpy 018, Vpy 022 [ 40 ], Vkat3, and Vkat2 (Yamashiro et al, unpublished,) were scored. These 10 loci were amplified in three multiplex reactions (Additional file 1 : Table S2) .…”

“…For DNA extraction, we first homogenized a 1-2 cm 2 piece of leaf with a TissueLyser and then subjected it to a silica-based purification method modified from Elphinstone et al (2003). We diluted DNA 1:16 and genotyped samples with 15 microsatellite markers ( Vince-02, Vince-04, Vince-06, Vince-08, Vince-11, Vince-14, Vince-15, Vince-23, Vince-26, Vince-31, Vince-32, Vince-39, Vinc-101, Vinc-102 and Vinc-118; Tada et al 2009, Supporting information). To improve the microsatellite peak profiles, we added a GTTT-tail to the 5′ end of each non-labelled primer (Brownstein et al 1996).…”

“…Some of the Apocynaceae species are outcrossing species, while others are predominantly selfing. In general, Apocynaceae outcrossing species are known to have high genetic diversity [21][22][23][24][25]. Many Apocynaceae outcrossing species have inbreeding coefficients close to zero, which indicates an outcrossing predominance.…”
