Development of Novel pH-Sensitive Eudragit Coated Beads Containing Curcumin-Mesalamine Combination for Colon-Specific Drug Delivery

Abstract: This research aims to develop a drug delivery system that effectively treats colitis while administering curcumin/mesalamine by coating alginate/chitosan beads with Eudragit® S-100 to target the colon. Beads were tested to determine their physicochemical characteristics. Coating with Eudragit® S-100 prevents drug release at a pH of less than 7; this was demonstrated by in-vitro release conducted in a medium with gradually varying pH to mimic circumstances in various regions of the gastrointestinal tract. “…A modified solvent kneading technique was used to prepare CURC and OMP inclusion complexes with HPR-β-CYD, assuming a 1:1 drug-to-cyclodextrin molar ratio [ 28 ].Accurate weights of either CURC or OMP were physically blended with HPR-β-CYD in a glass mortar adopting the geometric dilution technique. A solvent mixture of equal portions of chloroform, acetone, ethanol, and water (1:1:1:1) was added gradually with continuous stirring until a clear homogenous solution was formed.…”

“…The complexation of curcumin and HP-β-CD was achieved through solvent evaporation methods, as described in a previous report [43,44]. Curcumin and HP-β-CD were dissolved in a 1:2 ratio in ethanol and stirred at 40 β • C until the ethanol completely evaporated.…”
