Development of Sulfadiazine-Decorated PLGA Nanoparticles Loaded with 5-Fluorouracil and Cell Viability

Abstract: Abstract:The aim of this work was to synthesize sulfadiazine-poly(lactide-co-glycolide) (SUL-PLGA) nanoparticles (NPs) for the efficient delivery of 5-fluorouracil to cancer cells. The SUL-PLGA conjugation was assessed using FTIR, 1 H-NMR, 13 C-NMR, elemental analysis and TG and DTA analysis. The SUL-PLGA NPs were characterized using transmission and scanning electron microscopy and dynamic light scattering. Additionally, the zeta potential, drug content, and in vitro 5-FU release were evaluated. We found that… Show more

“…The method for culturing the A431 cell line has been described elsewhere [25]. Briefly, the A431 cells were cultured in DMEM medium with 10% FBS, l-glutamine (2 mM), penicillin and streptomycin (100 IU/mL), and HEPES (25 mM) and maintained at 37 • C in a humidified incubator containing 5% CO 2 .…”

“…Poly(lactic acid) (PLA) as the most important of the PHAs is derived from 100% renewable resources and has emerged as early as the 1970s as a material for degradable surgical sutures [9]. Since then it has been extensively explored for further uses in the medical arena such as drug delivery systems [10,11,12] and orthopaedic devices [13], often as one component in copolymers, e.g., with poly(ethylene glycol) [14] or poly(glycolic acid) [15], another important representative of the PHA class.…”

“…PLGA nanoparticles (NPs) and MPs have been widely used to deliver small molecule drugs [ 21 ], antigens [ 22 ], and siRNA [ 23 ], and have been established as safe in humans due to their biocompatible and biodegradable nature [ 24 - 27 ]. Here, we employed PLGA MPs as a scaffold to co-display peptide-MHC molecules (target antigen) and anti-Fas mAb (apoptosis-inducing molecules) in order to generate antigen-specific cytotoxic MPs.…”
