Developmental Regulation of Elastin Synthesis

Abstract: The regulation of elastin production has been evaluated and compared in two systems in the developing sheep: nuchal ligament and lung. Absolute rates of production in explant culture were estimated by immunoprecipitation of newly synthesized elastin and determination of precursor pool specific activities. Lung elastin production increased about 2.5-fold while nuchal ligament production increased about 9-fold during the latter half of gestation. In comparison, the activities of elastin messenger RNA(mRNAE) of t… Show more

“…These sections were first incubated for 30 min at room temperature with rabbit antibody against sheep elastin at a dilution of 1:40 with phosphate-buffered saline (PBS), and washed three times with PBS. This antibody has been previously shown to be specific for elastin according to a number of criteria, which have been given in detail elsewhere (Shibahara et al, 1981;Davidson et al, 1981). The sections were then incubated for 30 min with peroxidase-labeled anti-rabbit-IgG goat IgG (Cappel Laboratories) a t 1:20 dilution, and washed three times with PBS.…”

“…As noted, skin also contains a small mount of elastin but which is very thermally stable [151], for example elastin can survive boiling for several hours with no apparent change, and does not need attention here.…”

“…In old animals, independent of the parameters used, elastin was significantly (2/?<0.001) reduced compared with that in younger ages. Incomplete knowledge of the mechanisms involved in the regulation of elastin synthesis 38 hinders the interpretation of these phenomena. The age-related modifications of the elastic component might be related to altered ratios among extracellular matrix proteins, such as collagen or glycosylated molecules 41 ; to intrinsic rearrangements of elastic fiber constituents as glycosaminogrycans, 42 lysyl oxidase, 43 and vitronectin 44 ; and finally, to the dramatic reduction of elastin mRNA expression.…”
