Developmentally and stress‐induced small heat shock proteins in cork oak somatic embryos

Abstract: The timing and tissue localization of small heat shock proteins (sHSPs) during cork oak somatic embryo development was investigated under normal growing culture conditions and in response to stress. Western blot analyses using polyclonal antibodies raised against cork oak recombinant HSP17 showed a transient accumulation of class I sHSPs during somatic embryo maturation and germination. Moreover, the amount of protein increased at all stages of embryo development in response to exogenous stress. The developmen… Show more

“…As shown by the two-dimensional immunodetection patterns, in the holm oak, the anti-QsHsp17.4-CI antibody recognized two sets of immunospots in the 17 and approximately 10 kDa regions, respectively. Similar twodimwnsional patterns have been detected in Q. suber from cork and wood tissues and also in mature somatic embryos and plantlets (Puigderrajols et al 2002;Jofré et al 2003). Polypeptides in the 17 kDa region were induced by heat stress, whereas polypeptides in the 10 kDa region were specifically induced by oxidative stress (Puigderrajols et al 2002;Jofré et al 2003).…”

“…Developmental induction of Ha HSP17.7 G4 during zygotic embryogenesis was faithfully reproduced in the transgenic plants (Coca et al, 1996). Puigderrajols et al (2002) reported a transient accumulation of class I sHSPs during cork oak somatic embryo maturation and germination. This was localized to early differentiating, excluding the highly dividing regions of the root and shoot apical meristems.…”

“…There have been numerous studies concerning the hormonal induction of SE in a wide range of species. A significant amount of literature on auxin biosynthesis, metabolism, and transport in embryos has grown out of extensive analysis which shows that auxin plays important roles both in induction of embryo formation in culture and in the subsequent elaboration of proper morphogenesis during embryo development [42,149]. One of the well-established functions of the plant ubiquitin/ proteasome pathway is in auxin signaling as illustrated in Fig.…”
