The valorization of byproducts from the sugarcane industry represents a potential alternative method with a low energy cost for the production of metabolites that are of commercial and industrial interest. The production of exopolysaccharides (EPSs) was carried out using the yeast Suhomyces kilbournensis isolated from agro-industrial sugarcane, and the products and byproducts of this agro-industrial sugarcane were used as carbon sources for their recovery. The effect of pH, temperature, and carbon and nitrogen sources and their concentration in EPS production by submerged fermentation (SmF) was studied in 170 mL glass containers of uniform geometry at 30 °C with an initial pH of 6.5. The resulting EPSs were characterized with Fourier-transform infrared spectroscopy (FT-IR). The results showed that the highest EPS production yields were 4.26 and 44.33 g/L after 6 h of fermentation using sucrose and molasses as carbon sources, respectively. Finally, an FT-IR analysis of the EPSs produced by S. kilbournensis corresponded to levan, corroborating its origin. It is important to mention that this is the first work that reports the production of levan using this yeast. This is relevant because, currently, most studies are focused on the use of recombinant and genetically modified microorganisms; in this scenario, Suhomyces kilbournensis is a native yeast isolated from the sugar production process, giving it a great advantage in the incorporation of carbon sources into their metabolic processes in order to produce levan sucrose, which uses fructose to polymerize levan.

Aim: This study was aimed at assessing the effect of time interval between harvesting and processing on the physical and chemical quality parameters of pre-harvest burnt sugarcane Study Design: An experimental research design was carried out that involve random selection of samples and sent to laboratory for analysis Place and Duration of Study: The study was carried out on July from 26th to 30th 2023 in Kilombero Valley located in the eastern part of Tanzania, between latitude 7 0 42'42'S and longitude 37 0 00'00'E. Methodology: The NCO 376 variety of sugarcane was used, that were cut below ground level, the tops removed, stalks tightened in bundles. The sample size was 15. each sample constituted of 30 burnt sugarcane stalks samples that were randomly collected and sent to laboratory for analysis after 24 ,72 and 120 hours of storage. On every respective storage time, the samples were scratched with knife to remove external contaminants, the stalks were then milled using two roller machines to extract juice. The juice was filled and sealed in sterilized and labelled bottles ready for laboratory analysis. The laboratory analysis data collected were coded and entered in excel sheet and analysed using Statistical Package for Social Science (SPSS) version 25. Results: The result shows the means values for pH, brix , pol, purity and weight decreased with increase in storage time whereas the means values for dextran, yeast and Leuconostoc increased with increase storage time The results revealed that purity values were satisfactory in all storage time levels as compared to SASTA standard purity values (97.0 – 98.0% ), whereas mean values for brix and pol stored for 120 hours of storage time was not satisfactory.as compared to SASTA standard values for brix ( 18 – 23%) and pol (14 – 21%) .Dextran concentration was high as compared to South African Sugar Terminals (SAST) that maximum requirement for dextran is 150 mg/kg, dextran level as the measure of microbial deterioration was high in 72 and 120 storage hours indicating microbial contamination. Conclusion: Farmers and processors should be well informed about the consequence of delayed processing burned sugarcane to avoid economic loss for farmers and processors.

Physicochemical Characterization of Dextran HE29 Produced by the Leuconostoc citreum HE29 Isolated from Traditional Fermented Pickle

In this study, lactic acid bacteria (LAB) strains were isolated from traditional fermented pickles, and among the identified strains, Leuconostoc citreum HE29 with a strong slimy colony profile was further selected to determine the physicochemical and techno-functional properties of its exopolysaccharide (EPS). Glucose was the only sugar monomer in the core unit of EPS HE29 detected by HPLC analysis, and glucan HE29 revealed 7.3 kDa of molecular weight. Structural characterization of glucan HE29 by 1H and 13C NMR spectroscopy analysis demonstrated that EPS HE29 was a dextran-type EPS containing 5.3% levels of (1 → 3)-linked α-D-glucose units. This structural configuration was also supported by FT-IR analysis, which also demonstrated the functional groups within the dextran HE29 structure. In terms of thermal properties detected by TGA and DSC analysis, dextran HE29 demonstrated a degradation temperature of around 280 °C, showing its strong thermal features. A semi-crystalline nature was observed for dextran HE29 detected by XRD analysis. Finally, AFM and SEM analysis revealed tangled network-like properties and web-like branched structures for dextran HE29, respectively. These findings suggest the importance of plant-based fermented products as LAB sources in obtaining novel EPS structures with potential techno-functional roles.
