Diabetes affects endothelial cell function and alters fibrin clot formation in a microvascular flow model: A pilot study

Abstract: Diabetes is a proinflammatory and prothrombotic condition that increases the risk of vascular complications. The aim of this study was to develop a diabetic microvascular flow model that allows to study the complex interactions between endothelial cells, blood cells and plasma proteins and their effects on clot formation. Primary human cardiac microvascular endothelial cells from donors without diabetes or donors with diabetes (type 1 or type 2) were grown in a microfluidic chip, perfused with non-diabetic or … Show more

“… 4) Microfluidics present better biomimetic performance. Endothelialized microfluidics are rapidly evolving as humanized screening platforms ( Albers et al, 2019 ; Jenny et al, 2020 ; Dupuy et al, 2021 ). Increasing evidences demonstrate their great potential in simulating the interplays between platelets and circulatory systems ( Zhang et al, 2021 ).…”

“…When connected with external syringe pumps, such simple microfluidic setting enables systematic investigation of shear dependent platelet adhesion ( 43 ) and aggregation ( 44 ). Additionally, vascular cell cultures can be incorporated; specifically, endothelialized microchannels can model the interaction between blood components and the vessel wall under physiological hemodynamic parameters ( 45 ), or even when subjected to proinflammatory or other endothelial diseased conditions ( 16 ).…”

“…Accordingly, dysfunctional shift of endothelial cells toward pro-thrombotic and pro-atherogenic states instigates severe impairment of COVID-19 on the cardiovascular system [51]. Diabetes, like obesity, is characterized by hypercoagulable state as well [114], featuring hypofibrinolysis and elevated levels of plasminogen activator inhibitor-1 (PAI-1) complement as well as increased platelet aggregation [115,116]. In patients with established T2DM, neutrophil extracellular traps (NETs), complexes of chromosomal DNA, histones, and granule proteins, were higher compared with healthy individuals [117,118].…”
