Diagnosing viruses by the rolling circle amplified synthesis of DNAzymes

Abstract: Circular DNA is used as a template for the amplified detection of M13 phage ssDNA by a rolling circle amplification (RCA) process that synthesizes DNAzyme chains, thus enabling the colorimetric or chemiluminescent detection of the analyte.

“…The amplified nucleic acid can also serve as much more than a landing site for a hybridizing probe. Cheglakov et al (140) used a molecular beacon to initiate RCA on a circular template encoding a deoxyribozyme that mimicked the activity of HRP. When the substrate ABTS 2− [2,2 -azinobis(3-ethylbenzothiazoline)-6-sulfonic acid] was added in the presence of H 2 O 2 , a colorimetric change from clear to green was observed.…”

“…Specifically, the G-quadruplex-hemin horseradish peroxidase (HRP)-mimicking enzyme has been extensively used as a catalytic label for the amplified colorimetric (Li et al, 2009;Zhu et al, 2011), electrochemical Wang et al, 2014b) or chemiluminescent (Gao and Li, 2013;Luo et al, 2012;Zhou et al, 2012) detection of DNA. By combination with hybridization chain reaction (HCR) to assemble long nanowires or rolling circle amplification strategy, G-quadruplex-hemin complex itself can achieve the signal amplification process efficiently (Cheglakov et al, 2007;Shimron et al, 2012;Wang et al, 2014a;Weizmann et al, 2006).…”

“…DNAzymes with peroxidase activity have recently attracted great interest in medicine, biology, and material sciences. Some applications of this class of DNAzymes include detection of specific DNA sequences, measurement of telomerase activity, determination of heavy metals, and detection of small molecules and proteins such as cocaine and thrombin [1,[7][8][9][10].…”
