Diagnosis and treatment of diffuse large B-cell lymphoma

Abstract: Diffuse large B-cell lymphoma (DLBCL) is the most frequently-occurring type of malignant lymphoma in the Western world. It has an aggressive natural history, with a median survival of less than one year if left untreated. Immunochemotherapy regimens, consisting of the anti-CD20 antibody rituximab typically in combination with cyclophosphamide, doxorubicin, vincristine and prednisone (CHOP), are currently the treatment backbone. Despite remarkable progress in improving patient survival, clinical outcomes are st… Show more

“…Cytological reviews were possible in 14 of 22 BCL and 11 of 14 TCL cases, suggesting a predominance of DLBCL and peripheral TCL. This observation accords with both previous veterinary literature, in which these cytological classifications were the most commonly recognized among multicentric BCL and TCL cases [51], [55], [56], and reviews of human NHL [2], [57]. The similar cytological appearance of the most common types of canine and human BCL adds weight to the notion that the canine disease could model NHL [11].…”

“…Diffuse large B-cell lymphoma (DLBCL) is the most common non-Hodgkin lymphoma among adults, accounting for 25-50% of cases [1,2]. DLBCL is a heterogeneous disease, with multiple morphologic variants and histological subtypes [1][2][3].…”

“…DLBCL probably arises via a stepwise process of somatic mutations, particularly chromosomal translocations involving oncogenes and, often, promoter regions of the immunoglobulin genes (10). …”
