Diagnostic Accuracy of FOBT and Colorectal Cancer Genetic Testing: A Systematic Review & Meta-Analysis

Abstract: Introduction: Colorectal cancer (CRC) is the second leading cause of cancer related death in the world after lung cancer. Early detection of CRC leads to improvement in cancer survival rate. In recent years, efforts have been made to discover a non-invasive screening marker of higher sensitivity and specificity. Fecal occult blood testing (FOBT) and genetic testing become alternative modalities to screen CRC in the population other than colonoscopy. The aim of this systematic review and meta-analy… Show more

“…The statistically significant p value was set to 0.0167 in a one-way analysis of variance (ANOVA). Levels of FIBA 202-210 and PF4 [54][55][56][57][58][59][60][61][62] (p = 0.0134) increased in late-stage CRC groups compared to the HC group (Figure 1). Levels of ITIH4 [429][430][431][432][433][434][435][436][437][438] (p = 0.036), APOE [198][199][200][201][202][203][204][205][206][207] (p < 0.0001), and CFAH [58][59][60][61][62][63][64][65][66][67] decreased in early-stage CRC groups compared to the HC group.…”

“…These selected peptides corresponding to differentially expressed WGA-captured plasma proteins were then analyzed using an Agilent 6470 instrument to examine the quality of signals on a triple quadrupole. We discovered that only two methylated peptides and four unmodified peptides satisfied a signal-to noise ratio (S/N) of >5, including ADLS*GITGAR (AACT 341-350 ), HITSLEVIK (PF4 [54][55][56][57][58][59][60][61][62] ), LALDNGGLAR (ITIH4 [429][430][431][432][433][434][435][436][437][438] ), LGPLVEQGR (APOE [198][199][200][201][202][203][204][205][206][207] ), QLEQVIAK (FIBA [202][203][204][205][206][207][208][209][210] ), and SLGN*VIMVCR (CFAH [58][59][60][61][62][63][64][65][66][67] ) (* indicates a methylated site, Table S2). Information on targeted transitions, optimized collisions, and fragmentors is summarized in Table S2.…”

“…Any diseases with intestional bleeding such as enteritis, diverticulitis, inflammation bowel disease (IBD) and so on may produce false positive; at the same time, if the colon neoplasm does not bleed, it will appear false negative. Therefore, a supplementary of iFOBT should be developed [21][22]. The detection of tumor molecular markers in stool is a valuable screening method for colorectal cancer.…”
