Diagnostic accuracy of noninvasive polymerase chain reaction testing for the determination of fetal rhesus C, c and E status in early pregnancy

Abstract: Objective The aim of the study was to determine the sensitivity, specificity and accuracy of noninvasive tests for the fetal rhesus CcEc (RHCE) alleles C, c and E in early pregnancy.Design A prospective clinical trial was carried out to evaluate diagnostic accuracy.Setting Women were recruited at four centres specialising in prenatal diagnosis. Peripheral blood and amniotic fluid samples were obtained and sent to a single laboratory for analysis.Sample A total of 233 tests (46 for C, 87 for c and 100 for E) we… Show more

“…Due to interference of massive maternal DNA, it is difficult to identify the odd fetal chromosome. Therefore, cff DNA in maternal plasma was usually used in determination of X-linked genetic disordors, fetal RhD status, and some monogenetic diseases [12][13][14][15] . In order to use cff DNA for NIPD, real "fetal-phase-specific" DNA markers, which are highly specific, universal, and independent of sex or polymorphic, are needed.…”

“…However, both patients never performed Rh-matched transfusion. Overall, it seems that RhCE-matched transfusion is simple and practicable in clinics in China compared to other countries such as France, Brazil, the US, or Thailand [17,24,27,37,38]. Moreover we would like to suggest that β-thalassemia patients in China should receive Rh subgroup-compatible blood from the beginning of transfusion therapy.…”

“…In the light of this, a few studies have analyzed circulating cffDNA in maternal plasma for non-invasive prenatal RHCE and KEL genotyping and have demonstrated accurate test results [50,51].…”
