Diagnostic potential of Mab‐based ELISAs for antibodies to non‐structural proteins of foot‐and‐mouth disease virus to differentiate infection from vaccination

Abstract: This paper summarises the development of monoclonal antibody (Mab)-based immunoassays measuring antibodies to non-structural proteins of FMDV to differentiate infection from vaccination. Of the three non-structural proteins 2C, 3C and 3ABC evaluated in this study, the polypeptide 3ABC was the most immunogenic. Three ELISAs for the detection of antibodies to 3ABC were developed. Two assays rely on the competition of test sera against either a anti-3A Mab or against antisera to 3ABC raised in rabbits and guine… Show more

“…By contrast, the FMDV non-structural proteins 3A and 3C could not be detected in any of the tissues from animals after 28 days post contact infection (Table 2, Figure 3) [10], [11]. Antibodies specific for 3A and 3C readily detected infected cells in FMDV vesicles and lymph tissue during the acute phase of infection and in infected BHK-21 cells, co-localising with FMDV capsid (Figure S11 to S13).…”

“…A PI of Ͻ50% was considered negative, and a PI of Ն50% was considered positive for recent exposure (within the previous 6 to 12 months) (7,14). More specifically, a PI value of Ն50% but Ͻ70% was considered a weak positive result, and a PI value of Ն70% was considered a strong positive result (36).…”
