Dielectric Properties of the Plasma Membrane of Cultured Murine Fibroblasts Treated with a Nonterpenoid Extract of Azadirachta indica Seeds

Abstract: Neem oil is a natural product obtained from the seeds of the tree Azadirachta indica. In this report, we investigate the alterations of the biophysical properties of the plasma membrane caused by treatment with the nonterpenoid fraction of neem oil that we defined as methanolic extract (MEX). The dose-response effect was evaluated and a MEX-dependent cytoxicity evidenced. The effect of MEX on the plasma membrane was studied by a well-established dielectric spectroscopy technique: electrorotation, which allows … Show more

“…The results discussed in this work show that HeLa tumor cells are sensitive to a lower dosage of MEX as compared to stabilized mouse fibroblasts 3T6. Also, the cell membrane appears to be the main target of the extract as shown by lipoperoxidation assays: this observation, in particular, validates and expands former results published by our laboratory [ 7 ].…”

“…One of the research lines developed in our laboratory is the study of the biological activity of different natural compounds [ 4 , 5 , 6 , 7 ]. In particular, we recently evaluated the effects of MEX in cultured mouse fibroblasts where we observed a cytotoxic effect induced by this methanolic extract obtained from the whole Neem oil.…”

“…The conductance G is informative of the membrane function in terms metabolism/ion-transport. Therefore, this simplified model is commonly accepted as a good tool to investigate the biological membrane conditions [6]- [12].…”

“…In our laboratory MEX showed a significant and differential cytotoxic action, with the cancer cells being more sensitive than the normal ones [ 18 ]. The main target of MEX is the plasma membrane which, after treatment with this extract, becomes more fluid without a substantial loss of its structural properties [ 19 ]. In addition, preliminary experiments performed in our laboratory suggest that MEX has also an antiviral activity (Berardi et al ., in preparation); in any case a similar activity of neem leaf extracts was reported in a model of Dengue virus [ 20 ].…”
